usage - "Yes, Please" vs "Yes, Of course"

I have heard people saying:

Que: Can I use your pen?

Ans: Yes please.

and also

Que: Can I use your pen?

Ans: Yes Of course.

I wanted to know if there is any difference between these two replies(may be of context) or can they be used interchangeably?
